Solid Phase Peptide Synthesis
Small amounts of peptides for e.g. haematology, immunology or oncology research and development are often synthesized on automatic Peptide Synthesizers. The advantage of this approach is that many peptides can be synthesized simultaneously. However a major drawback is the lack of flexibility during synthesis. Depending on the synthesis protocol, terminated or deletion (missing amino acids in the chain) peptides can be formed. Since these last impurities have similar properties, purification will be laborious if not impossible. The failure of a coupling step can only be discovered if afterwards Quality Control by e.g. LC/MS is carried out.
Ansynth Service B.V. produces peptides at laboratory scale (about 10 - 1000 mg) mainly using Solid Phase Peptide Synthesis. Peptides are built on a polystyrene resin stepwise from C- to N-terminus. Our manual synthesis approach allows us to control and optimize the attachment of every amino acid and to estimate the coupling yield. After each step eventually remaining free amino functions of the growing peptide chain are terminated by means of an anhydride. Possible synthesis-related impurities in the crude peptide are mainly terminated sequences and almost no deletion sequences.
After detachment from the resin and removal of the side chain protecting groups the crude peptide can be purified. Dependent on the needs of the customer and the properties of the peptide a suitable target purity can be established. Most times, using preparative HPLC, crude peptides can be purified up to > 95% by HPLC.
For quality control of the synthesized peptides HPLC's with UV/Visible Light Detectors, and LC/MS methods are available.
In case a higher amount of peptide is demanded, it is preferred to first synthesize a small amount, e.g. 10 or 25 mg. This way information is obtained about synthesis problems, yield, and properties of the peptide regarding the requested purity. Based on data of the pilot synthesis a fair quotation for the intended amount can be proposed to the customer. The customer also has the opportunity to test the peptide before ordering a larger quantity.

RGD-Peptides
In recent years, the interest for the application of (cyclic) RGD-peptides in tumour-targeting has increased. As a result of this we have developed methodologies for the attachment of labels. For example cyclo[DdFKRG] and cyclo[DdFKRA] are wanted peptides and also the lysine-derivatives (e.g. Ata) are popular. As we prepare custom made peptides other novel variants are also possible, please inquire.
Confidentiality Agreement
Before we can present a quotation, we need to have specific information and at least the peptide sequence. Principally we consider all given information as confidential. But in general we can agree with the preliminary need of a Confidentiality Agreement between the Customer and Ansynth Service B.V.
Quotation, Prices and Delivery Time
In our opinion all synthetic peptides are special, so Ansynth Service B.V. has no standard amino acid coupling prices. Upon request we are pleased to send a specified quotation. The quoted price will depend on:
Synthetic aspects:

| expected difficulty in coupling steps |
| the presence of special building blocks (unnatural amino acids, amino acid derivatives) |
| the need for intra- or inter-molecular bridges (e.g. (cyclic) disulfides, head-to-tail cyclization) |

Purification aspects:
| required purity level e.g. crude, > 70%, > 80%, > 90% or > 95% on the basis of HPLC analysis |

| expected physical properties of the peptide (solubility) |
| the presence of one or more Cysteines in the sequence, and the wish of a (poly-)-S-S-cyclic peptide, or an S-S-dimer, or especially a peptide with free SH-functional groups |

Delivery Remarks
We prefer to start the synthesis at the beginning of the week. The given delivery time is based on the production time of an average peptide, but:
- We cannot guarantee a successful synthesis and purification in advance
- Whether we are capable to synthesize and to purify the peptide can only be shown in practice
- Due to complications during the synthesis and/or purification the delivery time can deviate, in this case the customer will be informed as soon as possible
- If the synthesis is successful but not the total ordered amount, we can partially deliver. Partial deliveries can be invoiced
- Information will be given about the delivery time of the complete order
- Shipping cost for deliveries to foreign countries will be partly charged as indicated in the quotation

We prefer to accept orders after a preliminary quotation. For preparing a quotation we need some information about the peptides to be synthesized:
